This is the nitty of how to do it manually, all examples being taken from a Microsoft Windows 2003 Server
Go to Start Menu/ Programs/ Accessories/ System Tools/ Backup
Click on the BACKUP tab and select where you want to save the backup to. In this example, it is a file on the local hard disk, located in directory AD_BAK: C:\AD_BAK\Backup.bkf
As in the example above, highlight and click "System State" You will see that Active Directory is more than one part, it is also the system state, boot files, registry etc (Maybe you now understand why I say have fun if you haven't made the backup prior to the crash. Depending on the number of servers, it may be easier to rebuild than try and join up again, especially if your Exchange is also split across servers). Click "Start Backup" and in my case, it took about 0.6Gb space for 15 users.
